Suspension of Amorphous Calcium Phosphate Nanoparticles Impact Commitment of Human Adipose-Derived Stem Cells In Vitro

چکیده
Amorphous calcium phosphate (aCaP) nanoparticles may trigger the osteogenic commitment of adipose-derived stem cells (ASCs) in vitro. The ASCs three human donors are investigated using basal culture medium DMEM to either 5 or 50 µg/mL aCaP suspension (control: no nanoparticles). After 7 14 days, cell marker genes, as well endothelial, osteogenic, chondrogenic, and adipogenic analyzed by qPCR. Free ion concentrations assessed supernatant. one week aCaP, downregulation markers ALP Runx2 is found, averaged across donors. Our results show that after two weeks, further downregulated, but upregulated. Endothelial such CD31 CD34, upregulated with a 2-week exposure. Inter-donor variability high: Two out significant upregulation at day compared aCaP. Notably, all changes obtained absence an medium. While chemical composition saturation status towards phases remain approximately same for conditions, gene expression considerably. Hence, potential endothelial ASCs.

Estrogen treatment enhances neurogenic differentiation of human adipose derived stem cells in vitro
Objective(s):Estrogen is a sexual hormone that has prominent effects on reproductive and non-reproductive tissues. The aim of this study is to evaluate the effects of estrogen on the proliferation and neural differentiation of human adipose derived stem cells (ADSCs) during neurogenic differentiation. Materials and Methods: Isolated human ADSCs were trans-differentiated in neural induction med...
متن کاملDifferentiation of Mesenchymal Stem Cells Derived From Human Adipose Tissue into Cholinergic-like Cells: In Vitro Study
Introduction: Cholinergic-associated diseases currently constitute a significant cause of neurological and neurodegenerative disabilities. As the drugs are not efficient in improving the suffered tissues, stem cell treatment is considered an effective strategy for substituting the lost cells. Methods: In the current study, we set out to investigate the differentiation properties of human adip...

متن کاملReprogramming by cytosolic extract of human embryonic stem cells improves dopaminergic differentiation potential of human adipose tissue-derived stem cells
The extract of pluripotent stem cells induces dedifferentiation of somatic cells with restricted plasticity. In this study, we used the extract of human embryonic stem cells (hESC) to dedifferentiate adipose tissue-derived stem cells (ADSCs) and examined the impact of this reprogramming event on dopaminergic differentiation of the cells. For this purpose, cytoplasmic extract of ESCs was prepare...
متن کاملExtract of mouse embryonic stem cells induces the expression of pluripotency genes in human adipose tissue-derived stem cells
Objective(s): In some previous studies, the extract of embryonic carcinoma cells (ECCs) and embryonic stem cells (ESCs) have been used to reprogram somatic cells to more dedifferentiated state. The aim of this study was to investigate the effect of mouse ESCs extract on the expression of some pluripotency markers in human adipose tissue-derived stem cells (ADSCs). Materials and Methods: Human A...
